Besides, histogram thresholding, a number of alternative approaches to image segmentation have been suggested in the literature, such as those based on feature space clustering, edge-detection, or watershed transformation. All these methods have advantages and disadvantages. We do not discuss these alternative methods, since the technique that we suggest in this paper is somehow more related to histogram thresholding than to the other approaches mentioned above. A comprehensive description of these approaches can be found in [7-11].

Effective segmentation of such biological images helps life-science researchers obtain useful imaging information for downstream analysis including disease diagnosis, treatment, and new drug discovery. The thresholding obtained from the minimization of the variograms is considered optimal in a spatial context, which can be combined with other optimal criteria for generalization or can be used for hysteresis thresholding as a hard-core threshold [1] by taking into account the information at various spatial distances.
